Examination on Abnormal Expression of Bax and Bcl-2 Proteins and Insulin Correction of Apoptosis in Hippocampal Subregions of Rats with Type 1 Diabetic Encephalopathy

Abstract

This study was aim to investigate the effects of insulin treatment on the expression of Bax and Bcl-2 proteins and
apoptosis of hippocampus cells in type 1 diabetic encephalopathy rats. In this paper, we will divide adult male rats
into three groups: encephalopathy group of type 1 diabetic, group of insulin treatment and group of vehicle model
treatment. To build an encephalopathy model of type 1 diabetic by injecting of streptozotocin at abdominal cavity,
then, injected 3IU of protamine long-acting zinc insulin into the model rat with encephalopathy subcutaneously to
establish an insulin treatment model. A vehicle treatment model was established by intraperitoneal injection of an
equal volume of citrate buffer and subcutaneous injection of an equal volume of physiological saline into healthy
rats. Morris water maze, Bax, Bcl-2 and TUNEL reaction techniques were used to observe apoptosis and protein
expression of Bax and Bcl-2 genes in hippocampal CA1, CA2 and CA3 subgroups of rats in each group. The
results showed that the number of Bax positive cells and TUNEL positive cells in the hippocampal CA1, CA2 and
CA3 sub regions of the water maze group were higher than those of the other groups (P<0.01) significantly.
However, the number of Bcl-2 positive cells in the hippocampal CA1, CA2 and CA3 subgroups of the
encephalopathy group was lower than that of the other groups (P<0.01) significantly. After treatment of the
encephalopathy group with insulin, the above indicators of the type 1 diabetic encephalopathy group were
reversed to near normal levels (P <0.01).The experimental results show that insulin therapy can correct the
expression levels of Bax and anti-apoptotic gene Bcl-2 in hippocampal CA1, CA2 and CA3 subtypes of type 1
diabetic encephalopathy rats.
